Winston-Salem is on Interstate 40 in the Piedmont, about an hour west of Greensboro and 90 minutes northeast of Charlotte. The nearest international airport is Piedmont Triad International (GSO) in Greensboro, about 25 minutes east by car. Charlotte Douglas (CLT) is the larger hub if you need international connections.
There is no passenger rail service to Winston-Salem itself — Amtrak's Carolinian and Piedmont services stop in Greensboro and High Point. Greyhound runs intercity buses. Within the city a car is the easiest way to get around; the Old Salem, downtown and Reynolda districts are too spread out to walk between in a single visit.
