Mestre is small and flat, so walking and cycling are the easiest ways to get around. The town has decent bicycle paths. For Venice, public buses cross the Liberty Bridge to Piazzale Roma in about 15 minutes, and the train from Mestre station to Venice Santa Lucia takes just 10 minutes and costs a couple of euros. Taxis are available but significantly more expensive. If you want to explore towns beyond Mestre, a rental car from the airport is the most practical option -- but cars are useless for Venice itself.
