Venice is the obvious day trip and barely qualifies as one -- the train takes ten minutes. But Mestre is also well placed for exploring the Veneto more broadly. Treviso, a charming walled town with canals of its own, is 30 minutes north by train. Padua, home to Giotto's Scrovegni Chapel and one of Europe's oldest universities, is about 25 minutes to the west.
